Several years ago, Dr. George Church and his team of Harvard scientists introduced a unique concept: DNA origami nanorobots. These nanoscale-robots, resembling clam shells, are constructed using synthetic DNA strands. They effectively enclose the drug payload until the designated time for administration. Unlike conventional controlled-release pills, the drug remains physically attached to the shell, preventing it from freely circulating within the body. The drug’s efficacy is solely dependent on the opening of the shell; closing it instantly halts the drug’s activity.

The nanobot’s entrance is regulated by a pair of locks, with each being a unique DNA strand known as an aptamer that attaches to a specific molecule, such as a receptor found on the exterior of cancer cells. Once the aptamer connects to the target, the clamshell opens, releasing the payload.

The Israeli team stated in their paper [a] that these “nanorobots represent a groundbreaking advancement in achieving precise control over therapeutic molecules”. However, a major challenge lies in identifying unique molecular targets found exclusively in diseased tissues for the nanorobots to interact with. This task is already daunting in the case of cancer, and becomes nearly insurmountable when dealing with mental disorders.

In order for DNA nanorobots to operate effectively under such circumstances, alternative methods of control are necessary.

This led the team to ponder: considering that mental disorders frequently exhibit distinct indications of irregular brain activity detectable through EEG (electroencephalogram), what if we employ brain waves to regulate DNA nanobots and the dispensation of medication?

Researchers injected different nanobots into cockroaches, labeling them with different fluorescent markers so they can follow and analyze how robot combinations affect where substances are delivered. The accuracy of this technique is similar to that of a computer system.

“This is the first time that biological therapy has been able to match how a computer processor works,” says co-author Ido Bachelet of the Institute of Nanotechnology and Advanced Materials at Bar Ilan University [b]. Unlike electronic devices, which are suitable for our watches, our cars or phones, we can use these robots in life domains, like a living cockroach,” says Ángel Goñi Moreno [c] of the National Center for Biotechnology in Madrid, Spain. “This opens the door for environmental or health applications.”
