The world has always been somewhat silent for those with hearing loss since birth. This condition, called congenital deafness, begins at birth and profoundly impacts a person’s communication, learning, and daily life. It is often due to genetic factors—changes in specific genes that are passed down through generations. Until now, the only options available in such cases were hearing aids or cochlear implants, which help but fail to fully restore natural hearing.
Meanwhile, a new study from the Karolinska Institute in Sweden has brought a ray of hope. This research, published in the prestigious journal Nature, suggests that gene therapy can significantly reverse a specific type of genetic deafness, especially in children and young adults.
In this study, scientists collaborated with Chinese hospitals and universities to study 10 patients aged 1 to 24. All of these patients’ hearing problems were due to mutations in the OTOF gene. This gene plays a crucial role in the production of a protein called otoferlin, which helps transmit sound signals from the ear to the brain. When this protein malfunctions, the ear can perceive sound, but the signals cannot reach the brain properly.
To address this problem, researchers used gene therapy. In this, a healthy gene is introduced into the body. For this, a safe virus, called adeno-associated virus, was used to deliver the functional OTOF gene directly into the ear. This procedure was performed through a small injection into the round window, a region at the base of the cochlea. The results were quite encouraging. Many patients began experiencing hearing improvements within just one month. After six months, all participants showed clear improvement. They could hear even softer sounds, previously impossible for them.
Notably, the greatest improvement was seen in children, especially those between the ages of 5 and 8. One young girl achieved near-normal hearing within a few months of treatment and was able to converse easily with her mother. This also indicates that if treatment is started early, results can be even better.
